一、服務內(nèi)容:

二、服務流程:
1. 客戶提交委托標記請求并明確標記要求,與公司商定具體的實驗項目、數(shù)量和費用,雙方簽訂《委托標記制備合同》。
2. 客戶提供待標記樣品,我公司按照合同要求安排標記服務,并在規(guī)定時間內(nèi)完成標記委托。
3. 標記完成后,我公司按照客戶要求分裝,免費低溫郵寄給客戶,同時提供標記報告。
三、樣品要求:
1. 待標記樣品需保證90%以上的純度,樣品中不含有干擾標記物結合的成分,如有特殊成分,請?zhí)崆皹俗⒄f明。
2. 待標記樣品最小標記量為1mg,最好是干粉,如果是液體,濃度應大于1mg/ml。
3. 抗體IgG樣品中應不含BSA、疊氮鈉、甘氨酸等成分。
4. 大分子蛋白應提供分子量、等電點、緩沖液成分及pH等信息;小分子多肽需提供氨基酸序列;小分子化合物還需提供分子結構式。
5. 請?zhí)崆罢f明任何特殊情況。

五、標記物介紹:
堿性磷酸酶(alkaline phosphatase,ALP或AP)是免疫診斷試劑產(chǎn)品最常用的標記酶之一,優(yōu)點是穩(wěn)定性高、靈敏度高。ALP是一種能夠?qū)孜锶チ姿峄拿?,即通過水解磷酸單酯將底物分子上的磷酸基團除去,并生成磷酸根離子和自由的羥基,這類底物包括核酸、蛋白、生物堿等。而該脫去磷酸基團的過程被稱為去磷酸化或脫磷酸化。堿性磷酸酶是磷酸酶的一種,磷酸酶的作用與激酶的作用正相反,激酶是磷酸化酶,可以利用能量分子,如ATP,將磷酸基團加到對應底物分子上。
辣根過氧化物酶(Horseradish Peroxidase,HRP)活性高,穩(wěn)定,分子量小,純酶容易制備,所以最常用。HRP廣泛分布于植物界,辣根中含量高,它是由無色的酶蛋白和棕色的鐵卟啉結合而成的糖蛋白,糖含量18%。HRP由多個同功酶組成,分子量為40,000,等電點為PH3~9,酶催化的最適PH因供氫體不同而稍有差異,但多在PH5左右。酶溶于水和58%以下飽和度硫酸銨溶液。HRP的輔基和酶蛋白最大吸收光譜分別為403nm和275nm,一般以OD403nm /OD275nm的比值RZ(德文ReinheitZahl)表示酶的純度。高純度的酶RZ值應在3.0左右(最高可達3.4)。RZ值越小,非酶蛋白就越多。
FITC和RhodamineB(RBITC)是常見的熒光素,性質(zhì)較穩(wěn)定;
Cy系列熒光染料由于熒光較強,對光猝滅有抵抗力而熒光不宜消退,近年來應用比較廣泛;
